چکیده

The Major Urinary Proteins (MUPs) of the house mouse, Mus musculus domesticus, are 19 kDa beta-barrel lipocalins that are involved in chemical communication between individuals. Many of them are excreted in urine where they play multiple roles, including coding of owner identity and transport, and slow release of bound volatile pheromones. It has been shown that mass spectrometric analysis of intact proteins, is capable of dissecting subtle structural differences between the members of this class of proteins. Moreover, mass spectrometric analysis of the intact proteins can contribute towards molecular phenotyping of MUPs .

Observation of heterogeneous gene products by FT-ICR MS.

The ability of FT-ICR MS to resolve isotopic variants of intact proteins for each of the charge states formed by electrospray ionization offers a sensitive, rapid method for detecting "low mass" heterogeneity, where this is defined as the presence of structural variants differing in mass by 2 Da or less. Such heterogeneity may reflect biological or chemical modifications of structure or may res...

Regulation of highly homologous major urinary proteins in house mice quantified with label-free proteomic methods† †Electronic supplementary information (ESI) available. See DOI: 10.1039/c6mb00278a Click here for additional data file.

Major urinary proteins (MUPs) are highly homologous proteoforms that function in binding, transporting and releasing pheromones in house mice. The main analytical challenge for studying variation in MUPs, even for state-of-the-art proteomics techniques, is their high degree of amino acid sequence homology. In this study we used unique peptides for proteoform-specific identification. We applied ...

Butylated hydroxytoluene is a ligand of urinary proteins derived from female mice.

Mice secrete substantial amounts of protein, particularly proteins called the major urinary proteins (MUPs), in urine. One function of MUPs is to sequester volatile pheromone ligands, thereby delaying their release and providing a stable long-lasting signal. Previously, only MUPs isolated from male mice have been used to identify ligands. Here, we tested the hypothesis that MUPs derived from fe...

Molecular heterogeneity in the Major Urinary Proteins of the house mouse Mus musculus.

Major Urinary Proteins (MUPs) from different inbred strains of mouse have been analysed by high-resolution ion-exchange chromatography and mass spectrometry. MUPs from six strains were resolved chromatographically into four major protein peaks which characterized two distinct phenotypes, typified by the profiles obtained from the Balb/c and C57BL/6 inbred strains. A combination of ion-exchange ...
